IN VITRO KOŞULLARDA KURAKLIK STRESİ ALTINDAKİ KOBER 5 BB ASMA ANACINDA BAZI FİZİKSEL VE BİYOKİMYASAL DEĞİŞİMLERİN BELİRLENMESİ

Abstract


Kuraklık stresinin asmaların bazı fiziksel ve biyokimyasal özellikler üzerine etkilerini belirlemek amacıyla gerçekleştirilen bu araştırmada bitkisel materyal olarak Kober 5 BB Amerikan asma anacı kullanılmıştır. In vitro bitkiciklerde kuraklık stresi yaratmak için besin ortamına %0, 1.2, 2.4, 3.6 ve 4.8 olmak üzere 5 farklı konsantrasyonda polietilen glikol (PEG) ilave edilmiştir. Farklı konsantrasyonlarda PEG içeren ortamlarda yetiştirilen in vitro sürgünlerde kültür sonrasında zararlanma derecesi, bitki ağırlığı, prolin miktarı, çözünebilir protein miktarı ile antioksidan enzim aktiviteleri (süperoksit dismütaz, katalaz ve askorbat peroksidaz) incelenmiştir. Artan kuraklıkla birlikte Kober 5 BB anacında büyüme ve gelişmenin gerilediği, stresin göstergesi olan biyokimyasal değişimlerde ise önemli farklılıkların olduğu belirlenmiştir. Diğer taraftan asmanın strese karşı koyabilmek adına bir takım mekanizmalar geliştirdiği; bunun sonucu olarak da bünyesindeki prolin ve serbest oksijen radikallerinin yok edilmesinde etkin olan SOD, CAT ve APX gibi antioksidan enzim aktivitelerini artırdığı tespit edilmiştir. Buna göre çalışmanın sonucunda SOD enzim aktivitesinin 14.74-32.21 ünite mg protein-1, CAT enzim aktivitesinin 5.64-12.04 µM dak-1 mg protein-1 arasında değiştiği belirlenmiştir. APX enzim aktivitesi ise 31.43-81.30 µM dak-1 mg protein-1 arasında değişim göstermiştir.
